Logic says that as long as you have pinout for ISP connection you can solder to mainboard. If there's no pinout available for the phone you need, then taking out the eMMC is the only way.
There's no "universal" pinout if that's what you meant and MTK phones that does communicate via USB can mostly be done via USB only so no changes here.
